IMPROVEMENT OF ROOTING OF CUTTINGS OF A DWARFING ROOTSTOCK FOR KAKI AND ITS MICROPROPAGATION

Abstract
We have found a promising dwarfing rootstock for kaki (Diospyros kaki Thunb.). The cuttings collected from its original tree rooted well, but the cuttings from the hedges propagated by cuttings did not root satisfactorily.
Therefore, we tried to improve rooting of the cuttings and investigated its micropropagation, because cuttings from hedges derived from micropropagation are known to root well.
Rooting of the cuttings taken from root suckers was better than that from the hedges.
Dipping the cuttings in 3000 ppm IBA solution for 5 s produced the best rooting of the other treatments tested, such as dipping in 0.5% IBA in talc, in 0.4% 1-naphthylacetamide in talc, and in 4% 3-hydroxy-5-methyl isoxazole in talc.
Dormant buds from the dwarfing rootstock were successfully established in vitro on MS medium supplemented with zeatin 5 μM. After multiplication of the shoots on the same medium, the shoots were dipped in 254 ppm IBA solution and were placed in rooting medium solidified with gellan gum and with and without vermiculite.
The addition of vermiculite to the rooting medium did not improve in rooting of the shoots and in acclimatization of the shoots with roots, although the shoots rooting in the medium with vermiculite had more secondary roots.
