EFFECTS OF ETHYLENE AND SILVER THIOSULPHATE ON VASE LIFE AND PROTEIN ELECTROPHORESIS OF OSMANTHUS FRAGRANS PETALS

Abstract
Osmanthus fragrans is one of the most famous traditional flowers in China.
The short vase life negatively affects its ornamental value and application.
This paper was conducted to study the effect of ethylene and silver thiosulphate (STS) on vase life and internal peptides.
The vase life was examined by comparing ethylene (25, 50, 100, 200 and 400 mg/L) and STS (0.2, 1 and 2 mM) treatment.
The results showed that 200 and 400 mg/L ethylene treatments could significantly shorten the vase life from 6.6 d (CK) to 3.6 and 3.3 d, respectively. 0.2 mM STS prolonged the vase life while 1 and 2 mM STS had the opposite effect.
We first established a SDS-PAGE system specially for testing Osmanthus fragrans Houban Jingui petals.
SDS-PAGE demonstrated that the amount of 39.8 and 27.5 KD peptides did not change and 37.6 KD peptides decreased gradually through the senescence process. 63.1, 57.5, 51.5 KD peptides appeared and increased along with the senescence process indicating certain correlation between peptides and senescence.
Ethylene probably accelerated petal senescence by regulating 51.5 and 32.3 KD peptides.
