CLONING AND BIOINFORMATICS ANALYSIS OF ERF RESPONSE FACTOR GENE FROM PRUNUS MUME

Abstract
Primers were designed according to the sequence of the ERF (ethylene responsive factor) genes from GenBank.
A fragment of the ERF response factor gene was isolated from Prunus mume by PCR and RT-PCR. The sequencing results showed that the sequences obtained by two approaches were the same.
The full-length DNA was 816 bp long, and encoded a putative protein of 271 amino acids.
On this basis, we analysed the structure and function of the sequence via bioinformation analysis using protocols and related software.
Sequences analysis showed that the gene owns the characteristics of the ERF response factor, which contains an AP2 /EREB DNA-binding domain and two special short amino acids sequences.
Homology analysis shows that the nucleotide were highly homologous to that of other species, especially the Malus×domestica. So we can predict that we have successfully cloned the ERF gene of Prunus mume. It will contribute to the follow-up downstream analysis and molecular breeding.
