In vitro establishment and proliferation of apple cultivars

Abstract
The goal of this study was to investigate in vitro multiplication protocols for the use with apple cultivars ‘Bohemia’, ‘Rubimeg’ and ‘Primadela’ grown in the Czech Republic.
For the initiation phase, dormant shoots (15-20 cm in length) were collected from field germplasm at Research and Breeding of Pomology Holovousy Ltd.
Shoot tips (5 to 10 mm in length) were successfully established in vitro using mercuric chloride at a concentration of 0.15% as a sterilization solution.
To assess the effect of plant growth regulators on multiplication, in vitro shoots were cultured on the basal MS medium supplemented with 1, 2 and 4 mg L-1 6-benzylaminopurine (BAP), 0.5 and 1 mg L-1 thidiazuron (TDZ), or 1, 2 and 4 mg L-1 kinetin.
BAP at the highest concentrations (2 and 4 mg L-1) and TDZ were found to be more effective than kinetin for shoot multiplication.
Generally, the highest proliferation rate was obtained for ‘Bohemia’ that produced 4.1 shoots (longer than 10 mm) on medium containing 4 mg L-1 BAP. On the contrary, the lowest proliferation rate (1.0) was obtained for ‘Rubimeg’ and ‘Primadela’, which almost did not multiply on MS medium containing the lowest concentration of BAP (1 mg L-1). Kinetin at the tested concentrations did not promote satisfactorily proliferation of the three apple cultivars (proliferation between 1.1 and 1.3). Efficient micropropagation protocols are a prerequisite for utilization of sophisticated in vitro virus elimination techniques (in vitro thermotherapy and chemotherapy).
