Abstract
Ballota acetabulosa (L.) Benth. is a bushy, evergreen xerophyte of the eastern Mediterranean region with potential use as a medicinal, ornamental and landscape plant.
In this study the micropropagation of the plant was investigated.
In vitro cultures were established from nodal explants, excised from in vitro grown seedlings, and cultured on MS medium supplemented with BA, zeatin, kinetin or 2iP at 0.5, 1.0 or 2.0 mg L-1. At the establishment stage, high shoot production (88-100%, 3-4 shoots explant-1) was achieved with 1.0 or 2.0 mg L-1 zeatin, 2.0 mg L-1 kinetin, or 1.0 mg L-1 BA. At the first subculture, higher shoot production was achieved from shoot tip explants when the medium was supplemented with either zeatin or 2iP or kinetin at 2.0 mg L-1 (87-100% response, 4-5 shoots per explant), while nodal explants responded satisfactorily only in the presence of 2.0 mg L-1 kinetin.
Almost all explants responded on hormone-free MS medium producing 1-2 shoots as well as roots.
All microshoots rooted in vitro on hormone-free MS medium or on MS medium with IBA at 0.5-4.0 mg L-1. All microplants were established successfully ex vitro in peat:perlite (1:1, v/v).
