Application of the DNA markers in Japanese tree peony cultivars

Abstract
Tree peony (Paeonia spp.) is a highly ornamental shrub of China origin and is one of the most popular woody ornamentals.
Shimane prefecture has the largest production volume of tree peonies in Japan.
The “Botan meikan” directory which cites 353 tree peony cultivars was published for curation of Japanese tree peony cultivars.
To establish a practical method to identify tree peony cultivars, we developed a method to distinguish tree peony cultivars in the “Botan meikan” using Random Amplified Polymorphic DNA (RAPD) markers.
On the comparison of RAPD band patterns between cultivar groups, we also developed two sequence tagged site (STS) markers.
The LuDeB marker able to detect the genetic relationship with P. lutea and P. delavayi in the French and American tree peony cultivar groups.
The HPB marker able to identify the cpDNA of herbaceous peony in inter-subgeneric hybrid cultivars between herbaceous and tree peony.
The possession of bands obtained in this study also supported the breeding history of cultivar development in the genus Paeonia. The utilization of the developed DNA markers is expected to improve curation of cultivars not only fields but also distribution, sales and gardens, and to open the way for new breeding using inter-specific hybrids and inter-subgeneric hybrids of tree peony.
