Abstract
A method for micropropagation of Norwegian cultivars of red raspberry (Rubus idaeus) via meristem cultures was tested.
Preliminary research was done on 5 clones using three MS media, differing mainly in vitamin content.
Two of these clones, from the Norwegian breeding programme at Kvithamar Research Station (B3: Distad x Viking and 27/09: Veten x Viking), showed promising regeneration rates.
These were micropropagated using MS medium and the medium of Anderson, which differs mainly in the amount of macronutrients.
The hormone ratio and the time of introduction of the hormones to the cultures was varied.
The meristems were subcultured to fresh medium every 4 weeks, and growth started after one generation in culture.
After 2 ½ generations in culture (10 weeks), several leaves were formed, and multiplication started.
The average multiplication factor 3–4 (3–4 shoots formed from one shoot during one generation) was finally obtained.
Cytokinin levels were tested in concentration ranges from 0 to 3.0 mg/1, and showed no significant difference for the multiplication ratio between 0.5 and 2.0 mg/1. Shoot tips were moved to an elongation medium before rooting them in the greenhouse directly in peat.
During summer 1989, the plantlets will be moved outdoors for field tests on growth and phenological characters.
