FACTORS INFLUENCING THE REGENERATION OF EUCALYPTUS GRANDIS BY ORGAN CULTURE

Some of the factors which influence the rooting of nodes of post-seedling Eucalyptus grandis in vitro are discussed.
These include:

1. The brown exudate produced by the explant and which is detrimental to the future development of the explant.
   The exudate is shown to be of phenolic composition.

2. The environment of the parent plant.
   A temporarily enhanced rooting ability of the nodes was obtained immediately after the parent plant had changed environment.

3. The position of the explant on the parent plant.
   The most important factor is the choice of the explant with respect to the physiological age of the leaf on the node.
   This leaf should be at least two-thirds of the final size with full pigmentation and not senescing.

Problems encountered with the adaptation of this technique developed for E. grandis to several other species of Eucalyptus are discussed.
