GROWTH AND DEVELOPMENT OF ASPARAGUS PLUMOSUS SHOOT TIPS IN VITRO

Lateral shoot tips from young Asparagus setaceus (Kunth) Jessop (syn.
A. plumosus Baker) shoots were grown on a modified Murashige and Skoog medium. – Tips from 5 to 20 mm lateral shoots had significantly better growth and development, than tips from lateral shoots (2 mm) still covered by the leaf-scale. – The highest temperature at which the cultures continued to grow was 21°C. The growth was faster at 24°C, but here the cultures stopped to grow after 4 weeks.
At 30°C there was nearly no growth at all.
Temperatures below 21°C inhibited growth and there was no growth at all at 9°C. – There was no need for the auxins: IAA and NAA. – Cytokinin was needed for growth and development.
Without cytokinin the cultures died.
The cytokinins: kinetin, benzyladenine (BA), zeatin, isopentyladenine (2iP) and tetrahydropyranylbenzyladenine (PBA) were examined in the concentrations 0.2, 2 and 10 mg/l.
With 0.2 mg/l the shoots appeared normal, with 2 and 10 mg/l the shoots were abnormal and compact with long chladophylls and with calluslike clusters of shoots around the nodes.
PBA and zeatin were found to be the most effective cytokinins. – Few plants have been developed from shoot tip cultures.
Shoots grown on 10 mg/l PBA developed shoot-clusters, which were grown on medium with PBA (2 mg/l) and different IAA concentrations (0.02, 0.2 and 2 mg/l IAA). After 6 to 12 months plants with shoots, crownbuds and roots appeared.
