IN VITRO MICROPROPAGATION OF SEVERAL GRAPEVINE CULTIVARS FROM ROMANIA

Abstract
Grapevine is one of the most important crops, thus, it is necessary to obtain plant material in order to develop a germplasm collection of valuable cultivars.
By in vitro culture it is possible to obtain numerous identical plantlets that could be preserved in a germplasm collection or used for different purposes.
In order to obtain high number of plants it is necessary to establish proper conditions of in vitro culture.
The following cultivars were studied: ‘Brumariu’, ‘Radames’, ‘Italian Riesling’, ‘Muscat Ottonel’, ‘Burgund’, ‘Traminer’, ‘Feteasca Alba’ and ‘Feteasca Neagra’. In vitro cultures were initiated starting with shoots and shoot tips explants, leaves and berries.
Callus induction was also obtained in all cultivars.
Shoot explants regenerated plants on all media tested, but multiplication was generally low.
The best results were obtained on ‘Muscat Ottonel’ and ‘Brumariu’. The best evolution showed shoot tip explants.
Leaves and berries generated callus, plant regeneration was achieved on MS medium supplemented with BA (2.0 mg/L) + ANA (0.1 mg/L), but after 2 weeks, explants turned brown and died.
In vitro propagation of grapevine is difficult to achieve because culture loses viability after several subcultivations.
