MULTIPLICATION OF CRATAEGUS MONOGYNA BY IN VITRO CULTURE OF NODAL SEGMENTS

Abstract
Micropropagation of Crataegus monogyna Jacq. (hawthorn) was investigated.
Multinodal segments collected from shoot tips of C. monogyna plants growing naturally in the Mediterranean island of Sicily were used as primary explants to establish in vitro cultures.
Subsequently, a 2×5 factorial experiment with all possible combinations of 5 concentrations (0, 2.22, 4.44, 8.88 and 17.76 μM) of benzyladenine (BA) and 2 concentrations (0 and 2.46 μM) of indole-3-butyric acid (IBA), was used to determine the optimum growth regulator combination for shoot multiplication by single node segments.
Increasing concentrations of indole-3-butyric acid (IBA) ranging from 0 to 9.80 μM were tested to induce adventitious root development on microshoots.
The highest rate of axillary shoot proliferation was induced on Murashige and Skoog (MS) agar medium in the presence of 4.44 μM BA and 2.46 μM IBA. The highest rate of rooting was induced on MS agar medium containing 4.90 μM IBA. Eighty percent of the in vitro rooted plantlets survived after transfer to soil.
