ASSESSMENT OF PHLOROGLUCINOL EFFECT ON ROOTING OF TISSUE CULTURED PERSIAN WALNUT

Micropropagated shoots of three cultivars of Juglans regia (‘Sunland’, ‘Chandler’ and ‘Kerman 120’) were used for evaluating phloroglucinol effect on rooting parameters.
In proliferation phase, four concentrations of phloroglucinol (0, 0.5, 1.0 and 2.0 mM) were added to the multiplication medium and some growth parameters affected by phloroglucinol were measured.
Then rooting was induced in 10 ppm IBA for 10 days.
Induced shoots were transferred to the rooting substrates for in vivo and in vitro rooting.
In root expression phase, two types of rooting substrates (Jiffy-7 pots and vermiculite: 1/4 strength DKW mixture) were tested.
The results showed that the phloroglocinol effect on rooting is different in each cultivar.
Phloroglocinol at 0.5 and 1.0 mM increased the rooting percentage.
Vermiculite: 1/4 strength DKW mixture was a better substrate for rooting in all of the cultivars.
Implication of this method on production of micropropagated plants will be discussed in this paper.
