EFFECT OF OSMOTIC STRESS ON IN VITRO SHOOT CULTURE OF PEAS (PISUM SATIVUM)

Abstract
Growth of in vitro shoot cultures of eight pea genotypes (‘Graphis’, ‘Erbi’, ‘Janus’, ‘Hanka’, ‘Baccara’, ‘Hunor’, ‘2107’ and ‘M49060’) was examined under different osmotic stress in order to study their ability to tolerate this stress.
Six nodal explants were placed on 50 ml media without PEG (control) or on media with 2.5, 5.0, 7.5 and 10.0% PEG-600 as different levels of osmotic treatments.
Cultures were grown with 16 h illumination, at 105 μmol.m-2.s-1 PPF light intensity and 22°C for 4 weeks.
The multiplication rate, the length of shoots and the number of nodes were recorded and compared to those which were cultured on the control media.
Intensity of callus development was also observed in all treatments.
Our results showed that genotypes can be divided in at least 3 groups according to their ability to tolerate osmotic stress at the 5.0 and 7.5% PEG levels.
In our experiments ‘Graphis’, ‘Hunor’ and ‘Janus’ have shown the best osmoregulation capability, while ‘M49060’, ‘Erbi’ proved to be sensitive to osmotic stress, considering all results.
