Abstract
A genotype-independent transformation procedure has been developed for apricot cultivars.
Transformation of meristematic cells with high regeneration potential would allow leaving out the adventitious regeneration process, the bottleneck of most transformation methodologies in fruit trees.
Here, we described a transformation procedure that has allowed relatively high transformation efficiencies of four different apricot cultivars.
We have taken advantage of the special characteristics of the meristematic cells which, when the meristem is removed, are still able to reorganise a new “organising centre” and then a new meristem.
By transforming the remaining cells after removing axillary buds, we have been able to produce transformed shoots.
The method seems to be very consistent and important modifications in the regeneration media do not significantly affect regeneration percentages.
Transformed plants could be obtained with frequencies (based in PCR evaluation) ranging from 1 to 7.6% depending on the cultivar and selection pressure.
