IDENTIFICATION OF APPLE TREE CULTIVARS GROWING IN BELARUS USING SSR-MARKERS

Abstract
The level of genetic differentiation among 108 apple-tree accessions was investigated with SSR markers previously identified in Malus ×domestica Borkh.
The set of accessions included old, modern, introduced cultivars growing in Belarus and some Malus species.
By using 20 SSR markers, we found 330 polymorphic alleles with an average number of alleles of 16.5 per marker.
Polymorphism information content (PIC) varied from 0.68 to 0.94 with an average of 0.84. In order to determine the phylogenetic similarity and relationships among cultivars, cluster analysis was conducted.
Apple accessions were grouped according to their pedigree and their geographic origin.
Many modern cultivars of Belarusian breeding are close in genetic aspect to old cultivars traditionally grown in this area and belonging to these pedigrees.
Wild species of Malus formed an individual cluster.
Three marker pairs covering various genome regions CH01c06 and CH02c02b, CH02b12 and CH04h02, CH03d12 and SdSSR were selected and used in multiplex PCR for identification of apple-tree genotypes at the molecular level.
These studies allowed to identify 108 genotypes of 108 apple-tree cultivars and species.
