Abstract
Intergeneric hybridization was performed on a beautiful Thai orchid by crossing Dendrobium lindleyi Steud. with yellow flowers as the female parent and the pollinia of Vanda curvifolia (Lindl.) L.M. Gardiner with red flowers as the male parent.
The purpose of this study was to determine the success rate of pod formation, in vitro hybrid seed germination, and multiple shoot formation from protocorms for micropropagation.
The results showed 50% success rate in hybrid pod formation, with mature pods measuring approximately 2.63 cm in length and 0.45 cm in width after 162 days of hand-pollination.
The hybrid seeds contained embryos and were able to germinate into protocorms after 86 days of sowing on modified Vacin and Went (1949) agar medium supplemented with 10 mg L‑1 GA3, 150 mL L‑1 coconut water, 10 g L‑1 sucrose, 7 g L‑1 agar, and the pH of 5.2. To induce multiple shoot formation for micropropagation of the hybrid protocorms, the effects of BA (0, 1, 2, and 3 mg L‑1) and TDZ (0.3 and 0.5 mg L‑1) on Murashige and Skoog (1962) agar medium were investigated.
The highest number of multiple shoots (26 shoots protocorm‑1) was observed when the hybrid protocorms were cultured on medium supplemented with 1 mg L‑1 BA, followed by 2 mg L‑1 BA and 0.3 mg L‑1 TDZ, resulting in approximately 17 and 13.20 shoots protocorm‑1, respectively after 45 days of culture.
