Abstract
The study on the propagation of the ‘Man Nong Saeng’ mango using plant tissue culture techniques aimed to determine the most effective sterilization formula for tissue growth, identify the optimal culture medium for inducing shoot and root development, and conserve the genetic resources of this unique mango cultivar.
Explants used in the study included shoots, lateral buds from mother plants, and nucellus tissues extracted from the seeds.
The results showed that the most effective sterilization formula was 50% Clorox for 30 min, yielding an 80% survival rate.
This was followed by 40% Clorox for 20 min with a 40% survival rate, and 60% Clorox for 30 min with a 20% survival rate.
The formulas using 30 and 20% Clorox for 20 min resulted in 0% survival.
Tissue culture trials using nucellus tissues extracted from the seeds demonstrated successful development in both shoot and root induction experiments.
For shoot induction, the most effective culture medium was Y3 supplemented with 1.5 mg L‑1 BA and 1.5 mg L‑1 NAA, which achieved an 80% shoot induction rate with an average shoot length of 6.3 cm.
This was followed by Y3 + BA 1 mg L‑1 + NAA 1 mg L‑1, with a 60% shoot induction rate and an average shoot length of 0.56 cm, and Y3 + BA 2 mg L‑1 + NAA 2 mg L‑1, with a 20% shoot induction rate and an average shoot length of 0.2 cm.
Both Y3 alone and Y3 + BA 0.5 mg L‑1 + NAA 0.5 mg L‑1 produced no shoots.
For root induction, Y3 + BA 1.5 mg L‑1 + NAA 1.5 mg L‑1 was again the most effective, with a 60% root induction rate and an average root length of 4 cm.
This was followed by Y3 + BA 2 mg L‑1 + NAA 2 mg L‑1, with a 10% root induction rate and an average root length of 2 cm.
Y3 and Y3 + BA 0.5 mg L‑1 + NAA 0.5 mg L‑1 showed no root induction.
