Abstract
Currently aquatic plants are gaining more popularity in traditional aquariums and water gardens.
We developed an efficient and cost-effective method for rapid and high-quality shoot multiplication and in vitro rooting of Echinodorus osiris using shoot tip explants derived from mature plants.
Shoot tip explants were excised from 90-day-old plantlets and cultured on MS (Murashige and Skoog, 1962) medium supplemented with various concentrations of growth regulators namely 6-furfurylaminopurine (kinetin; 5.0, 7.0, and 9.0 mg L‑1), and thidiazuron (TDZ; 5.0, 7.0, and 9.0 mg L‑1). The highest frequency (90%) of multiple shoot formation with maximum number of shoots (7.0 shoots explant‑1), shoot length (20.67 mm), root induction efficiency (100%), number of roots (31.50 roots explant‑1), and root length (39.65 mm) was achieved on MS medium supplemented with 5.0 mg L‑1 kinetin.
Regenerated plantlets were transplanted into 3.5×3.5 cm black net plastic pots containing rockwool medium for acclimatization for 45 days.
The acclimatization of in vitro raised plantlets was successfully undertaken with 100% survival rate in greenhouse conditions.
The present in vitro propagation protocol would facilitate an alternative method for rapid and large-scale production of this important ornamental aquatic plant.
