Abstract
‘Hom Chaiya’ rice is a renowned native rice cultivar from Surat Thani province that was popular in the past.
However, its popularity has recently declined, resulting in decreased cultivation by farmers and putting it at risk of extinction.
This study aimed to investigate the efficient tissue culture propagation methods of this rice cultivar.
Seeds of ‘Hom Chaiya’ rice were surface sterilized with Clorox solution at concentrations of 10, 15, 20, and 25% for 10, 15, and 25 min.
The sterilized seeds were then cultured on MS medium sterilized by adding 0.2% Clorox solution.
The results showed that sterilization with 15% Clorox solution for 20 min yielded the best results.
After one month of germination, seedlings were transferred for acclimatization in commercial potting soil (Rasmi Pan Mai brand) and paddy soil from Ban Pai Phra, Phunphin District, Surat Thani Province for 4 weeks. ‘Hom Chaiya’ rice seedlings grown in paddy soil showed the highest survival rate (80%); while, those in commercial potting soil had a 50% survival rate.
Regarding seedling tiller diameter, seedlings grown in paddy soil had the largest size at 1.35 cm and reached the greatest height of 42.46 cm.
