Abstract
Cit-SAP is a salt-associated protein found to be expressed in citrus plants, such as Etrog Citron, Cleopatra mandarin and Shamuti orange.
Its expression is increased following irrigation with saline water.
An elevated level of Cit-SAP was also detected when salt-tolerant Citrus sinensis ovular culture cells were grown in the presence of NaCl, ABA or PEG (Ben-Hayyim, Faltin, Gepstein, Camoin, Strosberg and Eshdat, submitted for publication). The gene encoding for Cit-SAP, designated csa, was isolated from cDNA expression library that was constructed from the NaCl induced salt-tolerant citrus cells.
A polypeptide chain containing 167 amino acids (Mr = 18568 kDa) can be deduced from the nucleotide sequence of csa. Partial amino acid sequence of the protein, as well as that deduced from the nucleotide sequence of csa, revealed a considerable homology to mammalian glutathione peroxidases; csa is also highly homologous to clone 6P229 recently isolated from tobacco protoplasts.
The increased expression of Cit-SAP in cultured cells exposed to salt, ABA and PEG, and its homology to glutathione peroxidase, suggest a putative molecular linkage between biochemical processes induced by different stress conditions.
