Abstract
In vitro regeneration of two plum genotypes was investigated starting from leaf explants.
As optimum regeneration medium was established MS basal medium with addition of 2 % sucrose, 0,5 μM/l IBA and 7,5 μM/l TDZ. Shoots were produced in 46,25 % for cv.
Stanley and 31,43 % for cv.
Kjustendilska sliva.
By histochemicaly assaying GUS activity transient transformation frequencies were established respectively for Agrobacterium tumefaciens strains EHA101 – 54 %, C58C1 – 46 %, Aglo – 34 %. These experiments are the basic prerequisite for the application of Agrobacterium mediated PPV coat protein gene transfer.
