Abstract
Embryo, ovule and ovary culture were studied to obtain interspecific hybrids between B.oleracea and B. campestris using broccoli, Chinese kale, rape, tsai-hsin and strain’I-40′. At first, to understand the development stage of embryo for culture, development of embryo, ovules and pods were observed by scanning electron microscope.
And effects of plant growth regulator, sampling position and planting time in vitro were investigated.
And effects of plant growth regulator, sampling position and planting time in vitro were investigated.
Cross compatibility was observed in intraspecies.
Development of embryosac was not observed in 15 days after pollination (DAP). Heart-shaped embryo developed to immature embryo in 30DAP. Interspecific hybrids were obtained by embryo culture or ovule culture between B.oleracea and B. campestris. It is considered that 18≈36DAP embryos and ovules were suitable explants for inducing hybrids.
The growing of embryo in vitro was promoted by 0.1≈1.0ppm NAA and kinetin.
Hybrid plants were tested to research their characters such as plant form and cold requirement.
The plant growth characters of hybrids were intermediate between their parental lines, especially in leaf shape and flower color.
