PLANT REGENERATION FROM LEAF-DERIVED CALLI OF GENTIANS AND THEIR PROTOPLAST CULTURE

Abstract
In order to improve culture conditions in gentian, the effect of TDZ on shoot induction from leaf derived-callus was examined.
Calli were induced from leaf explants of Gentiana scabra in various combinations of 2,4-D and BA. The critical factor for callus formation was the presence of 2,4-D. Shoot regeneration response from the callus was influenced by the kind of cytokinins in regeneration medium.
High frequency of shoot regeneration (100–80% of calli regenerating shoots) was obtained when calli were transferred to regeneration medium containing 0.5–1.0mgl-1 TDZ, while only 2.0–5.6% of calli induced shoots in the presence of 1.0–5.Omgl-1 BA. Plantlets were achieved by rooting from the shoots on a hormone-free MS medium.
High yield of protoplasts were enzymatically isolated from mesophyll cells of G. triflora and G. acaulis. Many colonies were formed from those of G. triflora on modified MS medium containing 2mgl-1 NAA and 0.5mgl-1 TDZ within 2 months of culture.
