Abstract
The
– glucuronidase(GUS) gene was introduced using electroporation and showed transient expression in leaf mesophyll protoplasts isolated from shoot cultures of ‘Fuji’ apple (Malus domestica Borkh.). The optimal condition for the transient expression of GUS gene was obtained at the electroporating in MES buffer(CPW solution, 0.6M mannitol, 5.0mM MES, 1.0% PVP) at 3 electric pulses of 200V/cm and 30ms.
The transient expression of GUS gene increased with DNA amounts treated up to 100μg/ml and 50μg/ml of salmon sperm DNA as a carrier DNA. This system could be used to test rapidly the efficiency of the promoters and the constructed genes in apple plant transformation.
