DISCRIMINATION OF CLONES OF VITIS VINIFERA L. BASED ON THE POLYMORPHISM OF STILBENE SYNTHASE-CHALCONE SYNTHASE 5′ UNTRANSLATED GENOMIC REGIONS

Many kind of molecular markers employed for cultivar identification of grapevine have been tested in the last years for discrimination of clones.
Unfortunately, they do not seem effective in discriminating the minimal differences, which may differentiate clones of some cultivars.
In our laboratory we are studying the multigene family that codes for the stilbene synthase-chalcone synthase enzymes in grapevine.
In particular we have cloned and sequenced the 5′ untranslated sequences and promoters of many putative members of such gene family.
Sequence analysis has revealed the presence of stretches of repetitive elements, some of which resemble true microsatellites.
After the establishment of a new method for varietal discrimination based on PCR amplifications using primers specifically designed to amplify such regions, we have tested the possibility of extension to clonal identification.
In this work we present preliminary data concerning the discrimination of clones of Chardonnay, Pinot noir and Sangiovese based on this method.
