Virus detection of cucumber tile leaf in Shandong Province

Abstract
An outbreak of cucumber (Cucumis sativus L.) virus disease occurred in Shandong Province from 2022 to 2023. It was called cucumber tile leaf by locals, and brought great economic losses to farmers.
In order to investigate the virus species, 31 samples were collected, and reverse transcription PCR was performed with 13 pairs of specific primers.
The results showed that watermelon silver mottle virus (WSMoV), cucumber green mottle mosaic virus (CGMMV), cucurbit chlorotic yellows virus (CCYV), tomato mosaic virus (TMV), and cucumber mosaic virus (CMV) were detected with detection rates of 80.65, 48.39, 19.35, 6.45 and 3.23%, respectively.
There were four types co-infections of WSMoV and CGMMV, WSMoV and CMV, WSMoV and CCYV, WSMoV and TMV. Co-infection of WSMoV and CGMMV, WSMoV and CMV were detected in Jiyang, with detection rates of 55.17 and 3.45%, respectively.
Co-infection of WSMoV and CCYV was detected in the Shouguang, with a detection rate of 6.90%. Co-infection of WSMoV and TMV has been detected in Shouguang and Jiyang, with detection rate of 6.90%. The results showed that cucumber tile leaves were mainly caused by WSMoV alone and its combination with CGMMV or CCYV. The severity of cucumber virus disease was exacerbated by nutritional and environmental factors.
