ZINNIA MARYLANDICA TISSUE CULTURE-INDUCED VARIATION AND HERITABILITY

Adventitious plantswere regenerated from Zinnia marylandica cotyledon tissue on Murashige-Skoog (MS) medium containing 0.2 or 22.2 μM thidiazuron (TDZ) and grown through flowering to evaluate for variation.
Plants derived from 0.2 μM TDZ had more variation than seed-derived control plants for seven of nine characteristics observed.
Plans derived from 22.2 μM TDZ had more variation than control plants for five of nine characteristics, and more variation than 0.2 μM TDZ-derived plants for two of nine characteristics.
Twelve of 149 0.2 μM TDZ-derived plants and three of 23 22.2 μM TDZ- derived plants that had variant characters were self-pollinated and progeny were observed for variation.
Aberrant characteristics, including plant height, fertility, flower color and morphology, were sexually transmitted in varying degrees from regenerated plants (Ro) to progeny (R1) indicating genetic change had occurred.
Additionally, aberrant characteristics not observed in regenerated plants arose in their progeny.
It is concluded that tissue culture regeneration of Z. marylandica plants by adventitious shoot formation on 0.2 μM TDZ- containing MS media successfully introduced heritable genetic variation.
