RESCUE AND CULTURE OF IMMATURE SEXUAL EMBRYOS IN TWO CROSSES OF ALSTROEMERIA (‘SAXONY’ X ‘TIARÁ’ AND ‘SAXONY’ X ‘AZULA’)

Abstract
Alstroemeria crop represents about 6% of the flower production in Colombia.
Commercially available varieties possess a wide diversity of colors and forms.
Breeding of Alstroemeria presents difficulties due to the genetic incompatibility associated with poliploidy that causes the abortion of immature embryos.
The objective of this research was to determine the optimal conditions for embryo rescue in early stages of development.
Two crosses were done using two types of Alstroemeria (‘Saxony’ x ‘Tiara’ and ‘Saxony’ x ‘Azula’). ‘Saxony’ and ‘Azula’ belong to the butterfly type and ‘Tiara’ is an orchid type.
Embryos grow normally up to nine days after pollination and after that, abortion ensues.
Therefore, embryos were rescued in the seventh day and placed on three different culture media, White, MS, and half-strength MS salts.
The best medium for the establishment was half-strength MS salts.
Oxidation was prevented with 50 mg/L of citric acid and 50 mg/L of ascorbic acid.
The best osmotic regulation and development of the embryos in both crosses, were obtained by a combination of 120 g/L of sucrose and 1 mg/L of gibberellic acid.
Plantlets were established on inert substratum under greenhouse conditions for their phenotypical evaluation.
