Abstract
Effect of different growth regulators, dark/light treatment, time of leaf excision, leaf age and repeated regeneration cycles on shoot regeneration from in vitro grown leaf explants was evaluated in clonal apple rootstock MM 106. A combination of BA and NAA was found to induce regeneration both directly as well as indirectly when kept in light.
First two apical and expanded leaves showed a greater regenerative ability along with more number of shoots/explant as well as longer shoots as compared to the 3rd and 4th leaves.
Leaf explants collected from 30 days old cultures resulted in higher regeneration frequency as compared to 20 days old cultures.
Repeated regeneration cycles showed that with the increase in number of cycles, regeneration percentage decreased but number of shoots per explant and length of shoots were found to be increased.
High multiplication rate (1: 10-12) was observed with regenerated shoots as compared to shoots from axillary bud cultures (1: 5-7).
