CRYOPRESERVATION AND PROTEOMIC ANALYSIS OF VANILLA (V. PLANIFOLIA A.) APICES TREATED WITH OSMOPROTECTANTS

Abstract
Apices isolated from in vitro plantlets of Vanilla planifolia Andrews were subjected to cryopreservation using a droplet-vitrification approach.
Survival (30%) and regeneration (10%) of new multiple shoots were achieved after 1 day pregrowth of apices on solid medium with 0.3 M sucrose, loading with 0.4 M sucrose + 2 M glycerol solution for 20-30 min and exposure to PVS3 for 30 min at room temperature.
This is the first report of regeneration of vanilla apices after cryopreservation.
For the proteomic analysis, we optimized the protein extraction method for vanilla apices.
Proteins from 1 day in vitro cultured apices on medium with or without PVS3 were compared using two-dimensional gel electrophoresis.
About 206 proteins were consistently reproduced from which 15 showed quantitative changes (13 increased the levels of their expression using the % volume criterion, and 2 decreased) in the PVS treated samples.
Qualitative changes were also detected in 2 proteins (1 induced and 1 repressed) of the same PVS samples.
