Abstract
In this study we tested a new cryopreservation method for conservation of old potato cultivars of the Czech origin.
Explants recovered after cryopreservation were introduced into ex vitro conditions and tests of genotype stability were evaluated by molecular markers, morphological traits and tuber proteins.
Fifty old potato cultivars of the Czech origin were cryopreserved and successfully recovered.
Genetic stability of cryopreserved cultivars was evaluated in 29 cultivars by molecular markers, 14 cultivars by morphological traits and 8 cultivars by tuber protein analyses.
Plants regenerated after cryopreservation were compared with plants from in vitro bank.
No significant differences were found between the samples before and after cryopreservation.
The results showed the possibility of using the cryopreservation method for long-term storage of potato germplasm.
