Abstract
The genebank Gatersleben and the Taxonomy department of the IPK maintain a large collection of Allium. In vitro culture is used for preservation of vegetatively propagated forms.
Sucrose-induced bulblets were stored at different temperatures.
Characterization of bulblet viability was performed by growth and ion efflux measurements.
Dormancy was compared in material stored at + 3°C with or without warm preculture at + 25°C and with or without treatments with ethylene.
Meristem culture was used for virus elimination.
Elimination rates depended on the clone and vary from 15 to 58%. Hybrids from distant crosses of onion with various wild species were obtained by embryo rescue via ovary culture.
They were verified by karyotype, isoenzyme, and RAPD analysis.
