Abstract
A liquid chromatographic method for the quantitative determination of free polyamines in bulbs of garlic is presented.
Polyamines were extracted with 5% HCIO4, stored at -20 °C and then converted to their benzoilated derivatives by the reaction with benzoyl chloride in alkaline medium.
The benzoilated samples were separated in a μBondapack C 18 (10μm), 3.9 mm ID x 30 cm column at room temperature using a 254 nm absorbance detector.
The solvent system consisted of methanol: water, run isocratelly at 64% of methanol, at a flow rate of 1.2 m1 / min.
Quantifications were carried out by the external standard method.
The calibration curves for the polyamines in the range of 0.2 – 3.4 umoles were lineal and sufficiently reproducible for quantitative determination.
Spermidine, cadaverine, putrescine and spermine were present in both the storage leaf and the inner sprout, which correlates well with the result of thin layer chromatography determinations.
