Abstract
The existence of retrotransposon-like elements in chrysanthemum was investigated using Dendranthema pacificum (Nakai) Kitam., a decaploid wild chrysanthemum species native to Japan.
Conserved sequences of reverse transcriptase domain of Tyl-copia group retrotransposons were used to synthesize primers for polymerase chain reaction (PCR). The size and sequences of amplified fragments were similar to that of corresponding region of Tyl-copia group retrotransposons.
This result suggests retrotransposon-like elements exist in D. pacificum. None of the sequences from three clones were identical.
One of the clones was more similar to the rice retrotransposon than to the other two elements from the chrysanthemum.
Heterogeneity of the sequences, activity of the elements, and possibility of horizontal transmission are discussed.
