ADVENTITIOUS SHOOT REGENERATION FROM IN VITRO CULTURED LEAVES OF APRICOT

Abstract
The effect of the physiological status of in vitro shoots, age of cultured leaves, growth regulator combinations, light intensity, gelling agents, length of induction period, on the percentage of regenerating leaves of the American apricot cultivar ‘Helena’ were studied.
The best regeneration results were obtained with leaves from proliferating shoots.
The regeneration of adventitious buds in these leaves was affected by cytokinin type and concentration, naphthaleneacetic acid (NAA) concentration and length of the dark period (optimum length of the dark period was 2-3 weeks). The highest regeneration percentages were obtained with thidiazuron (TDZ) (0.5 mg/l and 2 mg/l) and NAA (0.5 mg/l and 0.75 mg/l). benzyl-amino purine (BAP) induced very low regeneration percentages.
Agargel was a better gelling agent than agar or gelrite.
Reliability of results was high although the best percentage of regeneration obtained with this cultivar was only 35%. We are working to improve the efficiency of the developed methodology.
Important differences were found between cultivars.
With the apricot cultivar ‘Canino’ up to 60% of regenerating leaves have been obtained.
