Identification of ZjGULOs in Chinese jujube (Ziziphus jujuba Mill.) and their expression analysis in fruit development

Abstract
L-gulono-1,4-lactone oxidase (GULO, EC:1.1.3.8) is one member of the vanillyl alcohol oxidase (VAO) superfamily and belongs to aldonolactone oxidoreductase.
GULO was identified as the last step of AsA biosynthetic pathway in animals, whereas L-galactono-1,4-lactone dehydrogenase (GLDH) played this role in plants.
The fruit of Chinese jujube (Ziziphus jujuba Mill.) is rich in AsA content.
The GULOs’ contribution to AsA synthesis in jujube is largely unknown, thus identification, bioinformatics and expression of ZjGULOs were analyzed systematically.
In this study, seven ZjGULOs were first identified in the jujube genome, and most of which encoded acidic proteins.
Five ZjGULOs were located on close range of Chr. 8, indicating that tandem repeats happened in these genes.
Pedigree analysis also found that ZjGULOs existed in paralogous and orthologous events in the process of evolution.
Phylogenetic tree, gene structure and motif analysis showed that the GULOs were very conservative among different species, suggesting that they might play similar functions.
Promoter cis-acting element analysis showed that ZjGULOs could respond to light and drought stress, etc.
The protein interaction prediction showed that GULOs were likely to participate in AsA metabolism.
Further, qRT-PCR results showed the expression trends of ZjGULOs were different in jujube and wild jujube fruit, and most genes were highly expressed in half-red stage (S3) or full-red stage (S4). Thus, the results provided clues for the function study of ZjGULOs and would help to understand the relationship between GULOs and AsA metabolism.
