Optimization of simultaneous extraction of flavonoids, saponins and triterpenoid acid from jujube by response surface methodology

Abstract
Response surface methodology was used to optimize the simultaneous extraction of flavonoids, saponins and triterpenoid acids from ‘Luzao No.1’ jujube.
On the basis of single factor experiment, the extraction temperature, liquid-solid ratio and ethanol concentration were selected as independent variables, and the box-Benhnken central group method was used to design the three-factor and three-level experiment.
The response surface analysis was carried out based on the simultaneous extraction rates of flavonoids, saponins and triterpenoid acid as response values.
Design-Expert 8.0.6 software was used for multiple regression fitting of test data.
The results showed that the optimal extraction conditions of flavonoids, saponins and triterpenoid acid were as following: temperature 65°C, solid-liquid ratio 1:25 (g mL 1), ethanol concentration 70%. The simultaneous extraction of flavonoids, saponins and triterpenoid acid reached 4.34 mg g 1 under these conditions, which provided a theoretical basis for the deep development and comprehensive utilization of jujube resources.
