Effects of rainproof cultivation on organic acid accumulation related gene expression of Ziziphus jujuba ‘Dongzao’

Abstract
In order to study the effect of rainproof cultivation on the accumulation of organic acids in ‘Dongzao’ (Ziziphus jujuba Mill.), organic acid content in 12-year-old ‘Dongzao’ was determined by HPLC and organic acid-related genes were analyzed with assistance of transcriptomics.
The results show that: malic acid and quinic acid were the main acid components of the fruit in the full-red stage of ‘Dongzao’, and the content of main organic acids in the fruit was malic acid > quinic acid > citric acid > succinic acid.
Rainproof cultivation promoted the accumulation of malic acid, quinic acid, succinic acid and total acid at full-red stage of ‘Dongzao’ fruit and made the fruit flavor better.
Rainproof cultivation promoted the expression of positive acid metabolism regulation genes ZjPPC1, ZjPPC2, ZjCS1 and ZjCS2 in the genetic process, inhibited the expression of ZjpckA and DAHP synthase1 that negative regulating acid metabolism genes in the genetic process, and promoted the organic acid accumulation of ‘Dongzao’. The accumulation of organic acids in ‘Dongzao’ fruits was the result of the joint action of multiple enzymes.
