PRODUCTION AND CHARACTERIZATION OF POLYCLONAL AND MONOCLONAL ANTIBODIES AGAINST PEACH YELLOW LEAFROLL MLO-ASSOCIATED ANTIGENS

Abstract
Monoclonal (Mab) and polyclonal antibodies were developed to antigens from a mycoplasmalike organism (MLO) associated with the peach yellow leafroll (PYLR) form of peach western X (PWX) disease, originating in the Western USA. Three murine hybridoma cell lines secreting Mabs specific for the MLO antigens were obtained.
The polyclonal antibodies and all 3 Mabs reacted positively with sap extracts of Catharanthus roseus L. infected with the PYLR MLO and also with extracts of plants infected with the green valley X (GVX) isolate of PWX. No reactions were obtained with other fruit tree MLOs, including Molieres disease of cherry, apricot chlorotic leafroll, pear decline, and apple proliferation, nor were reactions obtained with several non-fruit tree MLOs.
The PYLR isolate reacted positively in F(ab’)2 tests with antibodies prepared to leafhopper-derived PWX antigens.
All three Mabs reacted positively with tissue extracts of PWX-infected plants and were used in field tests to detect MLO infections in Californian orchards.
No cross-reactions were obtained in reciprocal tests made using antibodies and antigens of PYLR MLO and of peach eastern X (PEX) MLO from New Jersey.

Polyacrylamide gel electrophoresis of preparations from PYLR-infected C. roseus followed by western blotting and immunoprobing with Mabs and polyclonal antibodies revealed a single major protein antigen of approximate Mr 29500 kDa.
