STUDIES ON THE DETECTION, TRANSMISSION AND DISTRIBUTION OF PEACH LATENT MOSAIC VIROID IN PEACH TREES

Peach latent mosaic (PLM) disease is induced by a viroid (PLMVd). The comparison between two diagnostic tests for PLMVd, a biological indexing procedure based in the cross-protection effect between different strains of this pathogen, and a biochemical technique of polyacrylamide gel electrophoresis (PAGE) specific for the detection of circular viroid-like RNAs, led to essentially coincidental results.
PAGE is more rapid but less sensitive than biological indexing.
Surveys using one or both of these approaches on peach orchards of the Valencia area showed that 88% of the trees of American varieties were infected by PLMVd.
Conversely, no PLMVd infection was detected in Spanish peach varieties which, however, presented 94% of infection (as revealed by PAGE) with a viroid of the hop stunt group apparently not associated with any disorder of the plants.
PLMVd was detected in different organs and plant parts of its host, with the only exception of the seeds.
PLMVd could be transmitted in the glasshouse by the aphid Myzus persicae.
