COMPARATIVE BIOLOGICAL, SEROLOGICAL AND ELECTROPHORETIC BEHAVIOUR OF SOME ISOLATES OF PRUNUS NECROTIC RINGSPOT ILARVIRUS

Abstract
A study was initiated to characterize the different symptomatological expressions caused by prunus necrotic ringspot ilarvirus (PNRV) in stone fruit species in Southern Italy and in Yugoslavia and to compare the serological and electrophoretic behaviour of different viral strains.
PNRV isolates, used in the comparative study, induced well distinguished symptomatologies in the field on different apricot, cherry, almond, peach, plum and rose varieties.

The biological behaviour of the virus isolates was investigated with field surveys, graft-transmission tests to almond, apricot, peach and plum indicators and mechanical transmission to a herbaceous host range made up of 24 species belonging to 7 different families.
The serological characterization was carried out by using gel double diffusion tests and ELISA, whereas the electrophoresis of purified preparation of different isolates was made in 2% agarose gel.

The isolates under study showed remarkable differences in their biological properties.
Serological differences among isolates were demonstrated in ELISA, but not in gel immunodiffusion tests.
Electrophoresis in agarose gel showed differences in the migration rates of intact viral nucleoproteins, which was also confirmed by immunoelectrophoresis tests.
