CHLOROPLAST DNA VARIATION IN SOUR CHERRY

Abstract
Restriction fragments of chloroplast DNA (cpDNA) from 29 sweet (Prunus avium L.), sour (P. cerasus L.), and ground (P. fruticosa Pall.) cherry selections and species hybrids were analyzed to determine the maternal origin of the tetraploid sour cherry.
Total leaf DNA was isolated and digested with two restriction endonucleases that preferentially cut cpDNA. The DNA fragments were then separated on agarose gels and filter-hybridized with two Pst I fragments of tomato cpDNA. Restriction fragment length polymorphisms (RFLPs) were identified that differentiated the cpDNA of sweet and ground cherry.
Of the 20 sour cherry selections, 17 had the cpDNA RFLPs of ground cherry while 3 had the cpDNA RFLPs of sweet cherry.
These results indicate that the allotetraploid sour cherry arose independently at least twice and that the more common maternal species is ground cherry.
