ANALYSIS OF TOMATO CULTIVARS AND HYBRIDS FOR IN VITRO CALLUS FORMATION AND REGENERATION

Abstract
Several tomato genotypes (cultivars and F1 hybrids) which differ in type of growth, yield and nutritional quality of the fruits, were studied for in vitro reactions.

This study was made in order to develop a procedure to regenarate plants with high frequency from apical meristems for each genotype.
Shoot apical meristems isolated from 7-day-old seedlings were aseptically cultured on a Murashige Skoog (M8) medium supplemented with varying concentrations and combinations of cytokinins and auxins under defined environmental conditions.
Benzyladenine (BA) at various concentrations (0.0225 – 2.25 mg/l) in conjunction with indoleacetic acid (IAA;0.175 – 1.75mg/l) and naphthaleneacetic acid (NAA; 0.0186 – 0.930 mg/l) were tested.
By using these phytohormones, individually or in combinations, we established 10 phytohormonal balances on which the reaction of 24 genotypes were studied.
The in vitro reaction was specifical and represented the result of the interaction between endogenous and exogenous levels of phytohormones.
Regenerated plants grew in soil to maturity and were found to be fertile and morphological true-to-type.
