DIFFERENTIAL EXPRESSION OF TOBACCO CHITINASE GENES IN RESPONSE TO VA MYCORRHIZAL FUNGI COLONIZATION

Symbiotic interaction between the vesicular arbuscular (VA) mycorrhizal fungus Glomuse intraradix and roots of tobacco (Nicotiana tabacum L. cv.
Xanthi-nc) was studied at the molecular level utilizing the differential display technique.
Total RNA from both mycorrhizal and non-mycorrhizal roots was extracted and cDNA was prepared and amplified by PCR. Forty bands were found to be different between the two treatments.
An amplified DNA band that was reduced in mycorrhitic as compared with non-mycorrhitic roots was extracted from the sequencing gel, reamplified, subcloned, and designated pMD17. Part of this clone showed 90–100% homology at the nucleotide level with the 3′ non-coding sequence of two basic chitinases from N. tabacum. Hybridization with a PCR amplified DNA fragment homologous to the 5′ end of tobacco basic chitinase gene and to a cDNA of tobacco basic chitinase showed the same hybridization pattern.
These findings suggest that this clone is part of the gene for basic chitinase and that its transcript was down-regulated at the early stages of mycorrhizal fungi colonization.
