Abstract
Hypoxis angustifolia var angustifolia corms were successfully placed into culture.
Initially both multiple shoots and callus developed from explants on the M&S basal medium without growth regulators.
Both direct and indirect plantlet regeneration was achieved by separating all meristematic tissue and undifferentiated callus at each subculture.
Maximum shoot production was obtained directly on M&S basal medium supplemented with 2 mg l-1 BA and indirectly on M&S basal medium supplemented with 0.5 mg l-1 BA. Since plantlet regeneration by organogenesis is difficult to achieve for corm producing monocotyledons, several distinct steps were introduced into the procedure.
Subsequent steps therefore included the sequential induction of corms and roots on separate M&S media, each without growth regulators.
Hardened off plantlets, transplanted to pots and acclimatized under high humidity conditions in a greenhouse at 30°C, flowered within six to eight weeks after transplantation.
