Abstract
Somatic embryogenesis was obtained from 3-week-old Eucalyptus nitens seedlings.
Explants were first cultivated for 3 days in a liquid medium containing a high concentration of auxin, then were transferred into 9 different media, containing a factorial combination of 3 concentrations – 0 – 0.5 μM – of both
-napthalenacetic acid (NAA) and benzylaminopurine (BAP). Seventy per cent of the genotypes produced globular somatic embryos after 6 weeks of culture in several NAA/BAP containing media.
Somatic embryos were obtained from hypocotyls, cotyledons, leaves and epicotyls with hypocotyls giving the highest proportion of embryogenic explant.
Hypocotyls also gave the highest number of embryo per competent explant.
After 9 weeks of culture in medium containing 0.5 μM BAP and 0.5 μM NAA, the globular somatic embryos began to proliferate by budding.
Embryogenic lines have been established and multiplied by adventitious embryogenesis and will be used in further experiments aimed at achieving maturation of the somatic embryos.
