METHODOLOGICAL AND GENETIC ASPECTS OF IN VITRO PLANT REGENERATION AND GENETIC TRANSFORMATION OF THE RECALCITRANT PEPPER (CAPSICUM ANNUUM L.)

Abstract
The transfer of the excellent regeneration capacity found in inbred line No. 40017 –13 was attempted in commercial, non regenerable pepper varieties by sexual crossing.
The obtained hybrids were genetically stabilized by the creation of dihaploid lines.
Results obtained in these experiments proved it is a way to improve the very poor regeneration ability of particular pepper varieties.
Genetic transformation experiments were carried out on cotyledon and rooted hypocotyl explants of the No 40017 R-13 inbred line using a disarmed Agrobacterium strain.
The key step in the Agrobacterium mediated genetic transformation of pepper using cotyledons as explants was the elongation of the shoots surviving the first kanamycin selection.
It was observed that cefotaxim used to eliminate the bacteria has a phytohormone effect detrimental to this regeneration step.
Interactions between antibiotics (cefotaxim, amoxycillin) and phytohormones (BAP, TDZ) during the regeneration process in cotyledon explants were revealed.
