Abstract
WRKY transcription factors (TFs) have been implicated in plant growth, development, and in response to environmental cues to regulate plant stress response and secondary metabolism.
However, there is limited information on WRKYs in Cymbidium sinense, which is an economically important species of flowering orchids cultivated in China, Japan, Korea, and Southeast Asia.
In this study, 52 CsWRKY proteins were identified in the transcriptome database of Cymbidium sinense and computationally analyzed for their physicochemical properties, conserved motifs as well as protein-protein interactions.
Sequence alignment and phylogenetic analysis classified these proteins into three major groups, namely I, II, and III with majority of WRKY proteins belonging to group II (30 CsWRKY), followed by group I (15 CsWRKY) and group III (7 CsWRKY). Group II proteins were further classified into 5 subgroups (IIa to IIe) based on their phylogeny.
Expression profiling of candidate CsWRKY genes in different plant tissues and in response to different plant hormone (abscisic acid, gibberellic acid, salicylic acid, and methyl jasmonate) suggested that most of the WRKY transcription factors were involved in multiple plant responses to hormone.
Among them, 4 CsWRKY (IIa1, IIa2, III1, III2) were strongly induced in response to various hormone treatments, indicating their potential important roles in hormone signaling.
Overall, our study represents the first survey of WRKY transcription factors in Cymbidium sinense, which could provide valuable information about the structural and functional characterization of CsWRKY genes.
