Abstract
Phytoplasmas (Mollicutes) are phloem-limited plant pathogenic bacteria.
They are transmitted by insect vectors from infected to healthy plants.
In our study, phytoplasma belonging to 16Sr-IX-H group causes abnormality in Brassica campestris. Phytoplasma-infected Brassica campestris showed flower abnormalities that were closely associated with changes in the expression of flower developmental genes.
The alteration in the expression of developmental genes and its association with DNA methylation was investigated as a possible mechanism.
Brassica plants were infected with phytoplasma by insect vector, Orosious orientalis, and grafting.
The study was carried out through RT-PCR, MSRE-PCR, Southern blotting, bisulfite sequencing, etc.
Transcriptional expression of methylated genes was found to be globally downregulated in plants infected with phytoplasma.
These results showed that genes particularly orthologous to Arabidopsis APETALAs involved in petal and stamen formation during flower development were downregulated severely in phytoplasma-infected plants.
The DNA methylation status of flower developmental gene in phytoplasma infected plants with 5-azacytidine restored gene expression suggesting that DNA methylation was involved in downregulation of floral development genes.
