In vitro response of calcium alginate-encapsulated shoot explants of Cistus creticus after short-term cold preservation

Abstract
Cistus creticus L. is an evergreen small shrub, heat-tolerant, native to the Mediterranean region, which has interesting medicinal and ornamental properties.
It blooms from late spring to early summer with many large single purple flowers, has low water requirements, and is increasingly used for landscaping.
The present study aims to investigate the creation of artificial seeds (synthetic seeds or synseeds) from in vitro-derived shoot explants and the preservation at 4°C for a period of up to 6 months to face further in vitro propagation.
By this way, the cost of in vitro subculturing is reduced; in addition, the in vitro plant material can be easily handled during transportation.
Prior to encapsulation, explants from in vitro cultures (shoot tips, 1st and 2nd node) were tested for their sprouting response in a solid MS nutrient medium enriched with 1 μM of BAP. Subsequently, combinations of sodium alginate (2.5%) with calcium chloride (50, 100, and 200 mM) were prepared for the encapsulation of the shoot tip explants and the formation of artificial seeds, which were then stored in a refrigerator at 4°C for 1, 2, 3, or 6 months.
The best treatment for the artificial seed formation was the combination of 2.5% sodium alginate and 100 mM calcium chloride.
This combination resulted in germination reaching 46.7 and 13.3% after 3 and 6 months of cold storage, respectively.
